초록/해제
요약Viral hemorrhagic disease (VHD) is a major public health concern with mortality rates ranging from 10-90%. While multiple virus families can elicit VHD, the mechanisms dictating the large ranges in fatality are not well understood. Both genetics and immune responses play pivotal roles in disease outcome. There are limited animal models of VHD. Many of the available models are expensive, require high biosafety containment, and/or extensively modified hosts or viruses. Herein, I describe a mouse model of VHD that accurately recapitulates human VHD symptoms without extensive genetic modification. This model is also safe and cost-effective. Furthermore, I have identified key immune cell populations and genetics that influence VHD susceptibility and pathogenesis. The results I have obtained here pave the way for both an improved animal model for studying VHD and insights into possible genetic candidates for therapeutic intervention.
