- 요약Direct cortical stimulation has been used for over a century to identify the role that specific brain areas play in perception and behavior. Within sensory cortex, this technique has been used to reveal the function of cortical regions, modulate perception, and provide sensory feedback as part of brain-machine interfaces. However, because of technological limitations, we have yet to understand how direct cortical stimulation impacts the neurons and neural circuits within sensory cortex. In particular, we have a poor understanding of how sensory cortex responds to artificial stimulation over time and how the evoked activity interacts with existing cortical circuitry. In this dissertation, I describe my work using optogenetics and optical imaging to study the dynamics of artificial sensory representations in mouse primary vibrissal somatosensory cortex (vS1). First, using a combined photostimulation and two-photon imaging approach, I observe neural activity across training as animals learn or fail to learn an optical microstimulation discrimination task. I show that training is associated with a sparsification of the photostimulus response and that animals with greater response stability are more likely to learn the task. Next, I compare activity evoked by optical microstimulation of vS1 with that evoked by natural sensory stimuli. I find that photostimulation engages recurrently coupled neurons responding to whisker touch more than expected by chance. Repeated exposure to simultaneous touch and optogenetic stimuli increases the interaction between the two stimulus representations. These results provide insight into how activity evoked by direct cortical stimulation interacts with existing cortical circuits and how variability in artificial sensory representations may influence perception and behavior.
