요약Lung squamous cell carcinoma (LUSC) is a major subtype of non-small cell lung cancer with limited treatment options. A distinct tumor-propagating cell (TPC) state that resembles normal basal cells has previously been identified within squamous tumors. We report that squamous tumors display unappreciated levels of intra-tumoral heterogeneity and harbor states distinct from the previously reported TPC state. We found that Krt13+ tumor cells represent a subpopulation of non-proliferative squamous tumor cells not found in other subtypes of lung cancer. Importantly, we discovered that KRT13 marks a hillock-like state in squamous tumors that displays signatures of immunomodulation. Importantly, we noticed that the hillock-like state is enriched in tumors initiated in club cells, consistent with normal developmental processes during regeneration of club cells. Within squamous tumors, we discovered that tumor-propagating cells can give rise to this newly identified Krt13+ hillock-like tumor cell subpopulation. This newly identified subpopulation may be regulated by KLF4 expression and highly produces the hillock-specific immunomodulator LGALS3. These findings provide important context into the tumor heterogeneity and plasticity displayed by squamous tumors.
