- 요약FSIAD, a type of female sexual dysfuction, is prevalent in menopausal women and women with depression who receive SSRI treatment. Current treatment options are limited for these clinical populations. Further, the role for central estradiol in the regulation of female sexual behaviors is poorly understood in mammals, however some evidence from the common marmoset suggests it may play a role in regulating these behaviors. To test the role of central estradiol in regulating sexual behavior, we designed a behavioral testing study followed by sn-RNA Seq analysis to determine hypothalamic targets of peripheral and central estradiol in mediating these behaviors. We established four treatment groups of female rhesus macaques to test this using experimentally replaced E2 and letrozole, an aromatase inhibitor: 1) E2+VEH, 2) E2+LET, 3) VEH, 4) LET. Females were tested at 10 mo and 14 mo after treatment onset with a previously acclimated breeding male, and at 18 mo were necropsied for hypothalamic collection. The results of this study reveal that both systemic and central estradiol are necessary for the complete demonstration of social behavior and female proceptive behavior, and that the combination of estradiol sources inhibit the expression of rejection behavior. Additionally, the sn-RNA Seq portion of this study identified several DEG in ESR1 highly expressing neurons that are of interest. Of particular interest is the higher expression of HTR2A and HTR2B in treatment groups that were less social and had more observed rejections in male paired behavior tests. These results support the current findings in the common marmoset and the mechanism of action for flibanserin, a treatment option for FSIAD. Our results also highlight additional targets of estradiol action in the hypothalamus that may inform clinical management of FSIAD.
