- 요약Mastitis, the inflammation of the mammary gland, remains a challenge in dairy production, often resulting from intramammary infections (IMI). While extensively studied in lactating cows, IMI also affects dairy heifers during pregnancy, contributing to reduced milk yields during first lactation. The objectives of this work were to characterize the impact of IMI on mammary tissue histology, determine whether IMI influence apoptosis and proliferation of mammary cells, and evaluate changes in collagen and elastin content in the mammary gland during late gestation. Collectively, these studies aimed to determine if, and in what ways, IMI alter mammary gland development in primigravid heifers. Twenty-one pregnant Holstein heifers were enrolled in the study. Contralateral, culture-negative quarters were randomly assigned to receive either an infusion of Staphylococcus aureus (CHALL) or sterile saline (SAL). Mammary secretion samples were collected at multiple time points for somatic cell count (SCC) analysis. Heifers were euthanized 21 days post-challenge, yielding tissue samples from animals at 6.5, 7.5, and 8.5 months of gestation. Mammary tissue was collected from two regions: the center parenchyma, located just above the gland cistern, and the edge parenchyma, adjacent to the mammary fat pad. Compared to SAL, CHALL quarters exhibited elevated SCC, increased tissue leukocyte infiltration, a nearly 50% reduction in luminal area in the edge region of 8.5-month pregnant heifers, smaller epithelial areas in the central region of 7.5-month pregnant heifers, and greater intralobular stromal areas in the same region and gestational stage. Additionally, CHALL quarters exhibited altered proliferation patterns during late gestation, with decreased mammary epithelial cell (MEC) proliferation in the edge region of 6.5-month pregnant heifers and increased stromal cell proliferation in the center region of 8.5-month pregnant heifers, compared to SAL quarters. Unexpectedly, CHALL quarters did not show increased MEC apoptosis. Instead, they exhibited greater stromal cell apoptosis across all gestational stages and mammary regions compared to SAL quarters. There was no evidence of increased collagen deposition in CHALL quarters. However, SAL quarters exhibited greater elastin abundance in the central region of the gland. Collectively, these findings indicate that IMI disrupt mammary tissue dynamics in a region- and stage-specific manner, rather than inducing uniform global changes. Such alterations during late gestation may impair mammary development and negatively impact first-lactation performance.
