요약The central dogma of molecular biology is the fundamental rule that explains how our genes are expressed, which allows for life to exist. The central dogma states that DNA is transcribed by enzymes known as RNA polymerases into RNA, which is then translated by cellular compartments called ribosomes, into protein, which carries out much of the functions our cells and bodies need. While RNA polymerases do make RNA from DNA templates, more needs to be understood about RNA polymerase activity that goes beyond the central dogma, including what they make in the absence of given DNA or in the presence of non-(intentional-)template DNA. The first half of the thesis discusses original scientific research discovering and characterizing activity of mitochondrial RNA polymerases that generate RNA monsters. Chapters 2 and 3 describe discovery and characterization of DragonRNA, a DNA→RNA combination molecule made by RNA polymerases when given short DNA oligonucleotide input. At the same time, while working to study the central dogma, genetics and biological research is being applied in many settings in our world, including two case examples: the fertility industry, and criminal justice and genetic forensics. The second half of this thesis focuses on applications of genetics and biomedical research in the law, and develops policy recommendations to make such applications more equitable and just. Chapter 4 discusses assisted reproductive technologies, fertility treatments to help people have genetic children, and how they currently serve to showcase gender and sexuality biases of our society, and makes recommendations and delves into science that can help combat those inequities. Chapter 5 reviews how genetic tools are used in criminal proceedings, including FBI-run databases and law enforcement use of private companies, and explains problems with genetic privacy, inequity, and inaccuracy in the current practices, and makes recommendations to improve accountability and oversight. This dissertation represents the work of an interdisciplinary PhD in Genetics studying monsters of many forms. Each chapter of this dissertation represents characterization of a different type of monster, from non-canonical nucleic acids to how genetics is applied in legal practice.
